Separation of T-even bacteriophage deoxyribonucleic acid from host deoxyribonucleic acid by hydroxyapatite chromatography.
نویسنده
چکیده
A simple and rapid method is described for separation of T-even bacteriophage deoxyribonucleic acid (DNA) from host (Escherichia coli) DNA by hydroxyapatite column chromatography with a shallow gradient of phosphate buffer at neutral pH. By this method, bacteriophage T2, T4, and T6 DNA (but not T5, T7, or lambda DNA) could be separated from host E. coli DNA. It was found that glucosylation of the T-even phage DNA is an important factor in separation.

Introduction: Low-energy helium-neon (He-Ne) laser beam lightis used in combination with sodium hypochlorite (Na2HOCl3) for clinical purposes. Regarding this, the present study aimed to investigate the effect of He-Ne laser (632.8 nm) and sodium hypochlorite on the calf thymus double-stranded deoxyribonucleic acid (ctdsDNA) molecule. Materials and Methods: For the purpose of the study, ctdsDNA...
متن کاملDeoxyribonucleic Acid Synthesis
Exposure of Escherichia coli cells to myxin results in the almost complete in'hibition of new deoxyribonucleic acid (DNA) synthesis, extensive degradation of pre-existing intracellular DNA, and a rapid loss of viability in these cells (9). After exposure to myxin for 30 min (<1% survivors and >25% degradation of DNA), infection of these cells by T4 bacteriophage results in the renewal of DNA sy...
متن کاملInhibition of host deoxyribonucleic acid synthesis by T4 bacteriophage in the absence of protein synthesis.
The requirement for phage protein synthesis for the inhibition of host deoxyribonucleic acid synthesis has been investigated by using a phage mutant unable to catalyze the production of any phage deoxyribonucleic acid. It has been concluded that the major pathway whereby phage inhibit host syntheses requires protein synthesis. The inhibition of host syntheses by phage ghosts is not affected by ...
متن کاملChange in methylation of Salmonella bacteriophage P3 deoxyribonucleic acid with host-controlled modification by Escherichia coli.
Modification of bacteriophage P3 by passage through Escherichia coli K was correlated with a 54% decrease in the content of 6-methylaminopurine in the phage deoxyribonucleic acid.
متن کاملSuperinfection with bacteriophage T4: inverse relationship between genetic exclusion and membrane association of deoxyribonucleic acid of secondary bacteriophage.
The majority of the deoxyribonucleic acid (DNA) of superinfecting T4 bacteriophage which is injected and not hydrolyzed does not attach to host cell membrane. Low levels of association of secondary phage DNA with membrane appear to be related to temporal genetic exclusion.
